I replaced the battery but the A320 still won't charge — is the new cell defective?

Not necessarily. The A320's charger expects a Ni-MH cell; if a different chemistry was fitted previously and damaged the charger's charge circuit, the charger may no longer deliver the correct voltage curve to the new Ni-MH pack. Check that the charger output is at or near 14.4V DC under no load with a multimeter. If the charger reads significantly below that, the charger is the fault — replace it before drawing any conclusions about the new battery.

The A320 battery faded fast — could leaving it on the dock have caused that?

Yes, continuous dock charging is one of the most common causes of early capacity fade in Ni-MH cordless vacuums. Ni-MH cells degrade faster under sustained low-level trickle charge than under normal charge-then-remove use. With the new battery, charge it fully and remove it from the dock — don't leave the A320 docked between every use.
